The Tool for Capturing Flag/HA/Myc-Tagged Proteins

Tag Antibodies Introduction

Tag antibodies are used for the detection and purification of tag sequences on various commercial expression vectors to analyze and determine the expression levels and functions of the target proteins. The principle behind their use is the antigen-antibody reaction, where these tag antibodies can specifically bind to the corresponding tag-fused proteins. Here is a brief list of some common protein tags suitable for use with tag antibodies:

Table 1. Introduction to Different Protein Tags

Tag

Sequence

Features

Applicable Scenarios

Flag

DYKDDDDK

1. Small molecular weight, does not change the function of the fusion protein, more hydrophilic than similar tags;

2. Contains enterokinase recognition site (DDDDK), which is convenient for tag removal when fused at the N-terminus of the target protein.

Suitable for purification and detection of low-expression level fusion proteins in eukaryotic expression systems.

HA

YPYDVPDYA

Derived from the hemagglutinin surface antigen determinant of the influenza virus; avoid using in experiments related to apoptotic cells.

Suitable for protein purification and detection.

Myc

Glu-Gln-Lys-Leu-Ile-Ser-Glu-Glu-Asp-Leu

Derived from the human proto-oncogene Myc; avoid using in experiments related to human cells.

Generally not used for protein purification, suitable for detection.

Yeasen tag antibody affinity purification products are made from high-quality mouse IgG monoclonal antibodies as raw materials. Through a process called "covalent coupling," these antibodies are directionally and efficiently coupled to agarose matrices or the surfaces of nano-magnetic beads. This not only reduces non-specific protein binding but also achieves a higher antibody load on the medium. Additionally, it provides tag antibody purification products suitable for various purification requirements, including different tag antibody purification resins and immunomagnetic beads, facilitating the purification of tagged proteins and immunoprecipitation experiments using manual or automated methods.
