The use of zebrafish Danio rerio as a model for intestinal damage is well established and the dextran sodium sulfate (DSS)-induced zebrafish IBD model mimics different features of mammalian IBD phenotypes, including neutrophilic inflammation in the intestine, excessive mucus production, increased intestinal lymphangiogenesis and upregulation of pro-inflammatory cytokines and markers ccl20, il1β, il23, il8, mmp9 and tnf-α[1].

1.3 Protocol
- Zebrafish embryos were obtained from natural spawning and raised until 1 day post fertilization (dpf) in an incubator at 28.5℃ in recirculating water (60 μg/mL instant ocean sea salts) and supplemented with methylene blue up to 1 dpf;
- After 1 dpf, use E3 embryo culture medium without methylene blue to culture to 3 dpf;
- Prepare 10% DSS storage solution with E3 medium;
- Dilute DSS with culture medium to the maximum non lethal dose (DSS concentration reference: 0.5%);
- The zebrafish was treated with 0.5% DSS from 3 dpf to 6 dpf, and the indexes were observed.
2 Results
The successful indicators of modeling: the number of neutrophils increased, acidic mucin increased, and the expression of inflammatory factors increased.
- Number of neutrophils: the number of neutrophils increased and moved to the intestine and epithelium;
- Acid mucus protein: It was verified by paraffin pathological section and AB-PAS staining method that the acid mucus protein in zebrafish intestine increased after DSS immersion;
- Inflammatory factor expression: The expression of inflammatory factor in zebrafish intestine increased after DSS treatment[1,2].
