It is widely acknowledged that traditional molecular diagnostic reagents present challenges in transportation and storage. To ensure the biological activity of effective components within the reagents (such as DNA enzymes, reverse transcriptases, etc.), they generally need to be stored and transported in a cold chain environment around -20°C. The requirement for cold chain transportation involves a large number of ice packs or dry ice, which is costly, and changes in ambient temperature can easily lead to repeated freezing and thawing of the diagnostic reagents, directly affecting the performance and shelf life of the reagents.
Lyophilized reagents offer multiple advantages, including the ability to be transported and stored at room temperature, ready-to-use upon arrival, and controlled transportation costs, making them the preferred choice for the development of molecular POCT (Point-of-Care Testing) or other novel molecular diagnostic reagents.
The selection of lyophilized raw materials is not merely about eliminating substances that cannot be freeze-dried. The selection process requires screening or adjustment for each component, and it is crucial to avoid blindly seeking materials that can be freeze-dried. Yeasen Biotech continuously increases investment in this technology and has developed a variety of high-quality lyophilizable raw materials and premixed liquid products. The company can also customize freeze-drying formulas for customers, helping them solve lyophilization challenges at the raw material level.
Advantages of freeze-dried products
- Reduce cost and operational complexity
- Support custom freeze-drying formula
- Provide reference freeze-drying process to assist the freeze-drying process
- After freeze-drying, the properties are consistent with liquid reagents, which can keep the enzyme activity intact
- Freeze-dried and still stable at room temperature, can be transported and stored at room temperature
The freeze-drying form is diversified, and the product has strong adaptability
Lyo-Ready Taq DNA Polymerase:stable at 37℃ for 54 days.
qPCR premixes were prepared using YEASEN 14316ES and corresponding glycerol-containing enzymes, respectively, and the template of 105, 104 and 103 copies/mL were amplified at the same time. The results showed that the amplification effect of glycerol-free 14316ES was consistent with that of glycerin-containing counterpart. The accelerated stability test of 13705ES was performed at 37℃. The results showed that the expansion performance of 13705ES was consistent with that of unaccelerated control after 54 days of accelerated treatment at 37℃. The above results showed that there was no difference in the performance of glycerol-free enzyme and glycerin-containing counterpart, and the stability was good.
Lyo-Ready qPCR Master Mix:One tube mix with freeze-dried protectant
Yeasen's lyophilized qPCR reagents are a one-tube mix (containing lyoprotectants) with no difference in detection performance compared to conventional qPCR mixes, and they support the detection of multiple pathogens. The reagents have excellent stability, with no significant difference in performance before and after lyophilization, and they perform well in lyophilization. Yeasen's lyophilized qPCR reagents (Cat#11893ES) were added to non-pig/HBV primers and probes and lyophilized into microchips, which were then compared with liquid reagents for performance testing, showing no significant differences.
Lyo-Ready RT-qPCR Master Mix:stable at 50℃ for 25 days.
Yeasen's lyophilized microarray shows no significant changes in Ct values, fluorescence values, or spherical shape before and after accelerated heat treatment, indicating good stability of the lyophilized product. RNA lyophilizable reagent (Cat#11831ES) was frozen into a lyophilized microarray and treated under different conditions (normal storage as T0, accelerated at 50℃ for 14 and 25 days), while amplifying SARS-CoV-2 pseudovirus templates at concentrations of 105, 104, and 103 copies/mL. The results showed no difference compared to the control.
The product range is diverse, supporting component separation
