Description
The T4 UvsY protein is a recombinant regulatory protein derived from the T4 bacteriophage. It plays a facilitating role in the homologous recombination process mediated by T4 UvsX Recombinase. During the search for homologous sequences, T4 UvsX Recombinase must compete with single-stranded DNA-binding proteins (SSBs) pre-bound to single-stranded DNA (ssDNA) for binding sites. The T4 UvsY protein enhances this competition, promoting the association of T4 UvsX Recombinase with ssDNA. With a molecular weight of approximately 16 kDa, T4 UvsY protein boosts the DNA-dependent ATPase activity of T4 UvsX Recombinase, reducing the minimum concentration required for its activity and facilitating strand displacement. Additionally, T4 UvsY protein aids T4 UvsX Recombinase in invading ssDNA-SSB complexes, leading to the displacement of SSBs and further enhancing the binding of T4 UvsX Recombinase to ssDNA. This enzyme exhibits no nuclease activity.

Primarily used in isothermal amplification techniques, such as Recombinase Polymerase Amplification (RPA).
