Description

The phosphorylation and dephosphorylation of proteins in cells are regulatory switches for many important biological activities such as signal transduction, cell proliferation, cell differentiation, and apoptosis. Therefore, controlling the action of phosphatases is of significant physiological importance in protein research. Tissues and cells are filled with a large number of proenzymes, which are fully activated after lysis and begin to digest surrounding proteins. Therefore, protecting the phosphorylation state of proteins is extremely critical in the study of protein phosphorylation pathways and signal transduction.

This product is a special mixture of multiple phosphatase inhibitors, which broadly inhibits non-specific acidic/alkaline phosphatases as well as specific protein tyrosine phosphatases, serine/threonine protein phosphatases (PPA, PP2A), etc.

The product is provided in the form of A and B tubes, and the two solutions can target different types of phosphatases to exert synergistic inhibitory effects.

Tube A contains Cantharidin, Bromotetramisole, and Microcystin-LR, provided as a ready-to-use solution in DMSO, inhibiting the L-isozyme of alkaline phosphatase and serine/threonine protein phosphatases, such as protein phosphatase 1 and 2A (PP1 and PP2A). Tube B contains Sodium orthovanadate, Sodium molybdate, Sodium tartrate, Imidazole, and Sodium Fluoride, provided as a ready-to-use solution in ddH2O, inhibiting tyrosine protein phosphatases, acidic phosphatases, and alkaline phosphatases.

This product can be used for WB, Co-IP, Pull-down assay, IF, IHC, kinase research, and various other analyses.

Shipping and Storage

The product is shipped with ice pack and can be stored at -15~-25 for 1 year, or stored at 2~8 for 2 months.

Instructions

Before use, take the product out of the refrigerator and allow it to warm to room temperature. Add A solution and B solution to the sample at a ratio of 1:100 (v/v), and mix thoroughly after each addition.

1. The solvent for Tube A is DMSO, and for Tube B is ddH2O. DMSO dissolves more slowly, so it takes longer to completely dissolve A solution compared to B solution. At lower temperatures, A solution can be placed in a 25water bath for dissolution.

2. Mixing A and B solutions before use can easily form precipitates, so this method should be avoided. It is recommended to add appropriate amounts of A solution and B solution separately to the sample.

3. This product can tolerate 1% SDS, Triton, NP-40.

Notes

1. It is recommended to aliquot and freeze the 100× stock solution to avoid repeated freeze-thaw cycles. Working solutions should be prepared fresh.

2. This product does not contain protease inhibitors. Appropriate protease inhibitors can be added to the sample to be analyzed according to experimental needs.

3. The solvent for Tube A is DMSO, please handle with care.

4. DTT, EDTA, EGTA, etc., may affect the sodium orthovanadate contained in this product, so handle with care..
