Flag polypeptide is a hydrophilic polypeptide encoding 8 amino acids (DYKDDDDK). It can be used to detect specific anti-Flag monoclonal antibodies. In order to improve the sensitivity of detection, 3×Flag repeated polypeptide (3× flag-tag Peptide) has been developed. This 3×Flag repeated polypeptide contains 23 amino acids and can detect fusion proteins up to 10 fmol. 3×Flag repeated peptides have been widely used to purify Flag fusion proteins and competitively bind Anti-Flag monoclonal antibodies.
The recommended working concentration of 3×Flag repeated polypeptide is 100-400 μg/mL.
