Description
The mitochondrial membrane potential fluorescent probe JC-10 is an upgraded product of JC-1 and can also be used to detect changes in mitochondrial membrane potential. Although JC-1 has been widely used in many experiments, it has poor water solubility and will precipitate in aqueous buffer solutions even at a concentration of 1 μM. JC-10 has better water solubility and can replace JC-1 in experiments that require high concentrations of dye.
In normal cells, JC-10 selectively aggregates in the mitochondrial matrix to form a reversible red fluorescent polymer (Ex=540 nm, Em=590 nm). In unhealthy mitochondria, due to the decrease or loss of membrane potential, JC-10 shifts from a polymeric form to a monomeric form in the cytoplasm, producing green fluorescence (Ex=490 nm, Em=525 nm). JC-10 can not only be used for qualitative detection, as the color change can directly reflect the change in mitochondrial membrane potential, but also for quantitative detection, as the degree of mitochondrial depolarization can be measured by the ratio of red to green fluorescence intensity. These two colors changes can be detected by standard filters on a flow cytometer, with green fluorescence analyzed by the FL1 channel and red fluorescence by the FL2 channel. In addition to flow cytometry, it can also be used for fluorescent imaging and fluorescence microplate detection platforms.
JC-10 performs better than JC-1 in some cell lines. However, the performance of JC-10 is highly cell-dependent.

Transported with wet ice. Store the powder at -20°C in a dry, light-protected environment for at least one year. Aliquot the storage solution into single-use volumes and store at -20°C in a dry, light-protected environment to avoid repeated freeze-thaw cycles, which is effective for about one year.
