Description
Nicotinamide, also known as Vitamin B3, is a non-competitive inhibitor of sirtuins, and its inhibitory effect affects a variety of important regulatory mechanisms in vitro and in vivo. Nicotinamide strongly inhibits yeast SIR2-mediated transcriptional silencing, increases rDNA recombination, and shortens replication lifespan. The yeast enzyme PNC1 converts nicotinamide to nicotinic acid, which activates Sir2 and prevents nicotinamide from telomere inhibition and rDNA silencing. Nicotinamide inhibits the deacetylation of p53 and inhibits the deacetylation of histones H3 and H4. Nicotinamide induces apoptosis in a variety of tumor cells, accompanied by caspase cascade activation.
Nicotinamide has been used in preclinical trials for the treatment of cancer and type 1 diabetes.
