Description
DAPT (GSI-IX) is a novel γ-secretase (γ-secretase) inhibitor that inhibits the production of total Aβ and Aβ42 in cells, which in turn affects cell signaling and cell differentiation. DAPT reduces the total Aβ yield in HEK 293 cells, with an IC50 of 20 nM in HEK 293 cells. DAPT inhibits the production of Aβ and Aβ42 in human neurocytes respectively with IC50 of 115 nM and 200 nM, which are 5-10-fold lower than those when applied to HEK 293 cells.
DAPT inhibits SK-MES-1 cell proliferation in a concentration-dependent manner with an IC50 of 11.265 μM. In addition, DAPT acts on lung squamous cell carcinoma cells and induces apoptosis-dependent and non-dependent apoptosis by inhibiting the Notch receptor signaling pathway. DAPT reduces lymphoid tissue proliferation in CBA-lprg mice and autoimmunity in MRL-lprr mice, and also exhibits resistance for several months in vivo, so DAPT has the potential to be an effective candidate in clinical trials for ALPS and lupus erythematosus (SLE).
