Description
MG132 is a potent, reversible, cell-permeable proteasome inhibitor (IC50=100 nM), more than 1000-fold more potent than ZLLal in inhibiting the ZLLL-MCA degradation activity of the 20S proteasome. MG132 also inhibits the cell cycle and calpain required for cyclin degradation (IC50=1.2 μM), and induces neurite outgrowth in PC12 cells. In addition, MG132 inhibited IκBα degradation, thereby inhibiting NF-κB pathway activation. MG132 induces apoptosis of MPM cells with the activation of caspase 3/7/8/9 proteins. In addition to inducing apoptosis, MG132 also induced changes in mitochondrial membrane potential and increased intracellular ROS levels.
