Description
Activin A is a member of the TGF-β family and has a wide range of biological activities, including regulation of cell proliferation and differentiation and promotion of neuronal survival. Elevated activin A levels in human colorectal and postmenopausal women have been associated with colorectal and breast cancer, respectively. The biological activity of activin A can be neutralized by inhibin and the diffusible TGF-β antagonist follicle suppressor. Activin A binds to two forms of activin receptor type I (action RI-A and action RI-B) and two forms (action RII-A and action RII-B). Activins are homodimers or heterodimers of different β-subunits. They are produced as precursor proteins with an amino-terminal prepeptide that is cleaved to release a C-terminal bioactive ligand.
This product is supplied as a lyophilized powder derived from HEK293 cells and is highly active and pure. Recombinant Human/Mouse/Rat Activin A is a 26.0 kDa disulfide bonded homodimer of two βA chains, each containing 116 amino acid residues.
Specification
Source |
Human Activin A Protein is expressed from HEK293 without tag. It contains Gly311-Ser426. |
Accession |
P08476 |
Molecular Weight |
The protein has a predicted MW of 12.97 kDa. Due to glycosylation, the protein migrates to 14-15 kDa under reduced (R) condition, and 23-25 kDa under Non reducing (N) condition based on Tris-Bis PAGE result. |
Endotoxin |
Less than 0.1 EU per μg by the LAL method. |
Purity |
> 95% as determined by Tris-Bis PAGE |
Activity |
Measured by its ability to inhibit proliferation of MPC-11 cells. The ED50 for this effect is < 7 ng/mL. |
Formulation |
Lyophilized from 0.22 μm filtered solution in 4 mM HCI. Normally 8% trehalose is added as protectant before lyophilization. |
Storage
-25 ~ -15 ° C storage, 1 year expiration date after receipt.
After re-solution, store at 2 ~8 °C, 7 days expiration date. After reconstitution, store at -85~-65 °C, 3 months expiration date.
It is recommended to aliquot the protein for the first use to avoid repeated freeze-thaw cycles.
Compounding Methods
Centrifuge before opening the cap. Use 4 mM HCl to reconstitute to a concentration of not less than 0.1 mg/mL.
