Description

CD7, also known as Leu-9, is an approximately 40 kDa glycosylated and palmitoylated transmembrane protein in the immunoglobulin superfamily. Mature human CD7 consists of a 155 amino acid (aa) extracellular domain (ECD), a 21 aa transmembrane segment, and a 39 aa cytoplasmic domain. Within the ECD, human CD7 shares 43% and 41% aa sequence identity with mouse and rat CD7, respectively. CD7 is expressed on T cells, NK cells, myeloid progenitor cells, and CD19+ B progenitor cells. Among CD8+ T cells, the CD7-bright population preferentially contains naïve and memory cells, while more weak expressors are primarily effector cells. CD7 associates in cis with CD3 and CD45 and in trans with SECTM1. Ligation of CD7 with antibodies or SECTM1 costimulates the activation and proliferation of CD4+ and CD8+ T cells and NK cells. It also enhances integrin-mediated adhesion of these cells to Fibronectin, ICAM-1, and VCAM-1 . CD7 additionally mediates the Galectin-1 induced apoptosis of T cells and facilitates the fusion of HIV-1 with target cells.

Recombinant Human CD7 Protein is expressed from HEK293 Cells with His tag and Avi tag at the C-terminal. It contains Ala26-Pro180.

Molecular Weight

Approximately 19.3 kDa. Due to glycosylation, the protein migrates to 34-48 kDa based on Tris-Bis PAGE result.

Purity

> 95% as determined by SDS-PAGE and HPLC.

Activity

ELISA Data: Immobilized Human CD7,His Tag at 0.5μg/ml (100μl/Well) on the plate. Dose response curve for Anti-CD7 Antibody, hFc Tag with the EC50 7.9ng/ml determined by ELISA.

Endotoxin

< 1.0 EU per 1μg of the protein by the LAL method.

Formulation

Lyophilized from 0.22 μm filtered solution in PBS (pH 7.4). Normally 5% trehalose is added as protectant before lyophilization.

Reconstitution

Centrifuge tubes before opening. Reconstituting to a concentration more than 100 μg/mL is recommended (usually we use 1 mg/mL solution for lyophilization). Dissolve the lyophilized protein in distilled water.

Storage

The product should be stored at -20~-80 for 1 year from date of receipt.

2-7 days, 2 ~8 °C under sterile conditions after reconstitution.

3-6 months, -20~-80under sterile conditions after reconstitution.

Recommend to aliquot the protein into smaller quantities when first used and avoid repeated freeze-thaw cycles.
