Description

Fc gamma RIIIa is a low/intermediate affinity receptor for polyvalent immune-complexed IgG. It is involved in phagocytosis, secretion of enzymes and inflammatory mediators, antibody-dependent cytotoxicity and clearance of immune complexes. In humans, it is a 50-70 kDa type I transmembrane activating receptor expressed by NK cells, T cells, monocytes, and macrophages. Fc gamma RIIIb is highly related, sharing 97% amino acid (aa) identity within the extracellular domain (ECD), but is a GPI-linked receptor expressed on human neutrophils and eosinophils. The ECD of Fc gamma RIIIa shares 63%, 61%, 65%, 59% and 58% aa identity with mouse Fc gamma RIV, rat Fc gamma RIIIa, feline CD16, bovine CD16 and porcine Fc gamma RIIIb paralogs, respectively. The Fc gamma RIIIa cDNA encodes 254 aa including a 16 aa signal sequence, 191 aa ECD with two C2-type Ig-like domains and five potential N-glycosylation sites, a 22 aa transmembrane (TM) sequence and a 25 aa cytoplasmic domain.

Source

Recombinant Human Fc gamma RIIIA/CD16a (F176) Protein is expressed from HEK293 Cells with His tag and Avi tag at the C-terminal.. It contains Gly17-Gln208(F176).

Molecular Weight

The protein has a predicted MW of 24.7 kDa. Due to glycosylation, the protein migrates to 48-58 kDa based on Tris-Bis PAGE result.

Physical Appearance

Sterile Filtered White lyophilized (freeze-dried) powder.

Purity

> 95% as determined by SDS-PAGE and HPLC

Activity

ELISA DataImmobilized Human Fc gamma RIIIA (F176) , His Tag at 2μg/ml (100μl/well) on the plate. Dose response curve for Anti-Fc gamma RIIIA Antibody, hFc Tag with the EC50 of 9.5ng/ml determined by ELISA.

SPR Data: Human Fc gamma RIIIA (F176) , His Tag captured on CM5 Chip via anti-His antibody can bind Rituximab, hFc Tag with an affinity constant of 0.92μM as determined in SPR assay.

Endotoxin

< 1.0 EU per 1μg of the protein by the LAL method.

Formulation

Lyophilized from 0.22 μm filtered solution in PBS (pH 7.4). Normally 5% trehalose is added as protectant before lyophilization.

Reconstitution

Centrifuge tubes before opening. Reconstituting to a concentration more than 100 μg/mL is recommended (usually we use 1 mg/mL solution for lyophilization). Dissolve the lyophilized protein in distilled water.

Storage

The product should be stored at -20~-80℃ for 1 year from date of receipt.

2-7 days, 2 ~8 °C under sterile conditions after reconstitution.

3-6 months, -20~-80℃ under sterile conditions after reconstitution.

Recommend to aliquot the protein into smaller quantities when first used and avoid repeated freeze-thaw cycles.

Note

1. Avoid repeated freezing and thawing.

2. Please operate with lab coats and disposable gloves, for your safety.

3. This product is for research use only.
