Description

Fibroblast growth factors (FGFs) comprise a family of at least eighteen structurally related proteins that are involved in a multitude of physiological and pathological cellular processes, including cell growth, differentiation, angiogenesis, wound healing and tumorgenesis. All four genes for FGF Rs encode proteins with an N-terminal signal peptide, three immunoglobulin (Ig)-like domains, an acid-box region containing a run of acidic residues between the IgI and IgII domains, a transmembrane domain and the split tyrosine-kinase domain. Additional splicing events for FGF R1-3, involving the C-terminal half of the IgIII domain encoded by two mutually exclusive alternative exons, generate FGF receptors with alternative IgIII domains (IIIb and IIIc).  Mutations in FGF R1-3 have been found in patients with birth defects involving craniosynostosis.

Recombinant Biotinylated Human FGFR1 alpha (IIIc) Protein is expressed from HEK293 Cells with His tag and Avi tag at the C-terminal. It contains Arg22-Glu374.

Molecular Weight

The protein has a predicted MW of 42 kDa. Due to glycosylation, the protein migrates to 60-80 kDa based on Tris-Bis PAGE result.

Purity

> 95% as determined by SDS-PAGE and HPLC.

Activity

ELISA DataImmobilized Anti-FGFR1 Anitibody, hFc Tag at 0.5 μg/mL (100 μL/well) on the plate. Dose response curve for Biotinylated Human FGFR1 alpha (IIIc) , His Tag with the EC50 of 25.9 ng/mL determined by ELISA.

Endotoxin

< 1.0 EU per 1μg of the protein by the LAL method.

Formulation

Lyophilized from 0.22 μm filtered solution in PBS (pH 7.4). Normally 5% trehalose is added as protectant before lyophilization.

Reconstitution

Centrifuge tubes before opening. Reconstituting to a concentration more than 100 μg/mL is recommended (usually we use 1 mg/mL solution for lyophilization). Dissolve the lyophilized protein in distilled water.

 

Storage

Transport in ice packs. Store at -20°C, valid for one year.

After reconstitution, store at -20 to -80°C in unopened state for 3-6 months. After reconstitution, store at 2-8°C for 2-7 days.

It is recommended to store in aliquots and freeze for the first use to avoid repeated freezing and thawing.
