Siglecs (sialic acid binding Ig-like lectins) are I-type lectins that belong to the immunoglobulin superfamily. They are characterized by an N-terminal Ig-like V-type domain which mediates sialic acid binding, followed by a varying number of Ig-like C2-type domains. Siglecs 5-11 constitute the CD33/Siglec-3 related group, and are differentially expressed in the hematopoietic system. Within the ECD, human Siglec-10 is most closely related to Siglec-5 (42% aa sequence identity). It shares 63% aa sequence identity with mouse Siglec-G. Siglec-10 is expressed on eosinophils, neutrophils, monocytes, and B cells with some splice variants predominating in particular cell types and tissue locations. It is upregulated on eosinophils in mouse models of allergic respiratory inflammation.

Molecular Weight

Activity

ELISA Data: Immobilized Human Siglec-10 at 2 μg/mL (100 μL/Well) on the plate. Dose response curve for Biotinylated Anti-Siglec-10 Antibody, hFc Tag with the EC50 of 14.4 ng/mL determined by ELISA.

Endotoxin

< 1.0 EU per 1μg of the protein by the LAL method.

Formulation

Lyophilized from 0.22 μm filtered solution in PBS (pH 7.4). Normally 5% trehalose is added as protectant before lyophilization.

Reconstitution

Centrifuge tubes before opening. Reconstituting to a concentration more than 100 μg/mL` is recommended (usually we use 1mg/mL solution for lyophilization). Dissolve the lyophilized protein in distilled water.

 

Storage

Transport with ice packs. Store at -20℃ to -80℃, valid for one year.

Once reconstituted, store unopened at -20 to -80°C for 3 to 6 months. After reconstitution, store at 2-8°C for 2-7 days.

It is recommended to store in aliquots and freeze for the first use to avoid repeated freezing and thawing.
