Description
Human Dendritic Cell-specific ICAM-3 Grabbing Non-integrin (DC-SIGN)/CD209 is a member of the C-type lectin family. The canonical DC-SIGN/CD209 isoform is a 46 kDa, 404 amino acid (aa) type II transmembrane protein. The extracellular region contains a Ca2+-dependent carbohydrate-binding lectin domain. Multiple human DC-SIGN/CD209 splice forms exist, generating both membrane-bound and soluble forms. DC-SIGN/CD209 is not well conserved between mouse and human, with the extracellular domain sharing only 63% aa identity. The DC-SIGN/CD209 lectin domain binds mannose oligosaccharides on pathogens including HIV as well as self glycoproteins including ICAMs. DC-SIGN/CD209 binds to butyrophilin 2A1 and this interaction can be blocked by HIV pp120. DC-SIGN/CD209 is expressed on dendritic cells (DC) and inflammatory macrophages and contributes to antigen presentation.

Source |
Recombinant Human CD209/DC-SIGN Protein is expressed from HEK293 Cells with His tag at the N-terminal. It contains Gln59-Ala404. |
Molecular Weight |
Approximately 40.5 kDa. Due to glycosylation, the protein migrates to 52-60 kDa based on Tris-Bis PAGE result. |
Purity |
> 95% as determined by SDS-PAGE and HPLC. |
Activity |
SPR Data: Human BTN2A1, His Tag immobilized on CM5 Chip can bind Human CD209, His Tag with an affinity constant of 0.12 μM as determined in SPR assay (Biacore T200). |
Endotoxin |
< 1.0 EU per 1μg of the protein by the LAL method. |
Formulation |
Lyophilized from 0.22 μm filtered solution in PBS (pH 7.4). Normally 5% trehalose is added as protectant before lyophilization. |
Reconstitution |
Centrifuge tubes before opening. Reconstituting to a concentration more than 100 μg/mL is recommended (usually we use 1 mg/mL solution for lyophilization). Dissolve the lyophilized protein in distilled water. |
Storage
Transported with ice packs. Store at -20℃ to -80℃, valid for one year.
After reconstitution, store unopened at -20 to -80°C for 3 to 6 months. After reconstitution, store at 2 to 8°C for 2 to 7 days.。
It is recommended to store in aliquots and freeze for the first use to avoid repeated freezing and thawing.
