Description

CD73, an ecto-5'-Nucleotidase, is an ectoenzyme expressed by most cell types. The 5'-Nucleotidase activity of CD73 converts extracellular nucleoside5'monophosphates to nucleosides, with AMP as the preferred substrate. CD73 is one of several enzymes responsible for the production of extracellular adenosine, a signaling molecule that is involved in responses to inflammation and tissue injury. CD73 is a lymphocyte maturation marker that has functions independent of its catalytic activity. These functions include the adhesion of B-cells to follicular dendritic cells and T-cell signaling. CD73 is also a regulator of leukocyte extravasation, a function that requires its 5'-Nucleotidase activity. The native enzyme is a homodimer bound to the cell membrane through a glycosyl phophatidylinositol (GPI) anchor.

Recombinant Human CD73/NT5E Protein is expressed from HEK293 Cells with hFC tag at the C-terminal. It contains Trp27-Lys547.

Molecular Weight

Approximately 84.5 kDa. Due to glycosylation, the protein migrates to 85-100 kDa based on Tris-Bis PAGE result.

Purity

> 95% as determined by SDS-PAGE and HPLC.

Activity

ELISA Data: Immobilized Human CD73, hFc Tag at 1μg/ml (100μl/well) on the plate. Dose response curve for Biotinylated Anti-CD73 Antibody, hFc Tag with the EC50 of 19.7ng/ml determined by ELISA.

Endotoxin

< 1.0 EU per 1μg of the protein by the LAL method.

Formulation

Lyophilized from 0.22μm filtered solution in 20mM Tris, 120mM NaCl (pH 7.5). Normally 8% trehalose is added as protectant before lyophilization.

Reconstitution

Centrifuge tubes before opening. Reconstituting to a concentration more than 100 μg/mL is recommended (usually we use 1 mg/mL solution for lyophilization). Dissolve the lyophilized protein in distilled water.


Storage

Transported with ice packs. Store at -20℃ to -80℃, valid for one year.

After reconstitution, store unopened at -20 to -80°C for 3 to 6 months. After reconstitution, store at 2 to 8°C for 2 to 7 days.

It is recommended to store in aliquots and freeze for the first use to avoid repeated freezing and thawing.
