描述
T4 多核苷酸激酶 (T4 PNK) 是一种 5'-羟基激酶，可促进 γ-磷酸从 ATP 转移到寡核苷酸或多核苷酸链（包括双链和单链 DNA 或 RNA）的 5'-羟基末端以及 3'-单磷酸核苷酸。这种转移是可逆的。T4 PNK 还具有 3'-磷酸酶活性，可使 3'-磷酸化的寡核苷酸末端、脱氧 3'-单磷酸核苷酸和脱氧 3'-二磷酸核苷酸去磷酸化。在 ADP 存在下，T4 PNK 可充当 5'-磷酸酶，促进 5'-磷酸化的寡核苷酸或多核苷酸上的 5'-磷酸基团与 ATP 的交换。该酶广泛用于构建 DNA 文库和末端标记以生成探针。
