描述
烟酰胺又称维生素B3，是一种非竞争性的sirtuins抑制剂，其抑制作用影响体内外多种重要的调控机制。烟酰胺强烈抑制酵母SIR2介导的转录沉默，增加rDNA重组，缩短复制寿命。酵母酶PNC1将烟酰胺转化为烟酸，烟酸激活Sir2，阻止烟酰胺抑制端粒和rDNA沉默。烟酰胺抑制p53的去乙酰化，抑制组蛋白H3和H4的去乙酰化。烟酰胺诱导多种肿瘤细胞凋亡，伴随caspase级联激活。
烟酰胺已在治疗癌症和 1 型糖尿病的临床前试验中使用。
