描述
MG132 是一种强效、可逆、可穿透细胞的蛋白酶体抑制剂（IC50=100 nM），在抑制 20S 蛋白酶体的 ZLLL-MCA 降解活性方面比 ZLLal 强 1000 倍以上。MG132 还抑制细胞周期和细胞周期蛋白降解所需的钙蛋白酶（IC50=1.2 μM），并诱导 PC12 细胞中的神经突生长。此外，MG132 抑制 IκBα 降解，从而抑制 NF-κB 通路激活。MG132 通过激活 caspase 3/7/8/9 蛋白诱导 MPM 细胞凋亡。除了诱导细胞凋亡外，MG132 还诱导线粒体膜电位变化并增加细胞内 ROS 水平。
