Description
CD40 Ligand, also known as TNFSF, a type II membrane protein with homology to TNF, is transiently expressed on activated T cells and known to be important for B cell Ig production and for activation and differentiation of monocytes and dendritic cells. Mature human CD40 Ligand consists of a 22 amino acid (aa) cytoplasmic domain, a transmembrane segment, and an 215 aa extracellular region . It is primarily expressed on activated CD4+ T lymphocytes, and also found in other types of cells, like NK cells, mast cells, basophils and eosinophils. In total CD40 ligand has three binding partners: CD40, α5β1 integrin and αIIbβ3. CD40 Ligand dysregulation on T cells and antigen presenting cells contributes to the immune deficiency associated with HIV infection and AIDS. CD40 ligation by CD40 Ligand promotes B cell activation and T cell-dependent humoral responses.

Source |
Recombinant Human CD40 Ligand/TNFSF5 Trimer Protein is expressed from HEK293 Cells with monomeric hFc tag at the N-terminal. It contains Met113-Leu261. |
Molecular Weight |
Approximately 74.9 kDa. Due to glycosylation, the protein migrates to 75-80 kDa based on Tris-Bis PAGE result. |
Purity |
> 95% as determined by SDS-PAGE and HPLC. |
Activity |
ELISA Data: Immobilized Human CD40, His Tag at 1μg/ml (100μl/Well) on the plate. Dose response curve for Human CD40 Ligand (Trimer) , hFc Tag with the EC50 of 0.13μg/ml determined by ELISA. Blocking Data:Serial dilutions of Anti-CD40 Antibody were added into Biotinylated Human CD40, His Tag : Human CD40 Ligand (Trimer) , hFc Tag binding reactioins. The half maximal inhibitiory concentration (IC50) is 2.0ug/ml. |
Endotoxin |
< 1.0 EU per 1μg of the protein by the LAL method. |
Formulation |
Lyophilized from 0.22 μm filtered solution in PBS (pH 7.4). Normally 5% trehalose is added as protectant before lyophilization. |
Reconstitution |
Centrifuge tubes before opening. Reconstituting to a concentration more than 100 μg/mL is recommended (usually we use 1 mg/mL solution for lyophilization). Dissolve the lyophilized protein in distilled water. |
Storage
Transported with ice packs. Store at -20℃ to -80℃, valid for one year.
After reconstitution, store unopened at -20 to -80°C for 3 to 6 months. After reconstitution, store at 2 to 8°C for 2 to 7 days.
It is recommended to store in aliquots and freeze for the first use to avoid repeated freezing and thawing.
