Wnt-3a belongs to the Wnt family of signaling proteins, which plays an important role in maintaining the integrity of embryonic and adult tissues.The Wnt family is highly conserved and cysteine-rich.The Wnt signaling pathway can be divided into classical and non-classical pathways, and is involved in the β-catenin-mediated classical signaling pathway including Wnt1, Wnt2, Wnt2b, Wnt3, Wnt3a, Wnt7a, Wnt8, Wnt8b and Wnt10a. Wnt3a, Wnt7a, Wnt8, Wnt8b and Wnt10a, the classical signaling pathway regulates stem cell pluripotent differentiation, organ development and regeneration.Wnt3a signals through the frizzled (FZD) family of receptors and the LRP5/6 co-receptor to regulate gene expression and a variety of cellular behaviors.Wnt3a, as the most representative signaling protein of the Wnt family, has a wide distribution, and has a role in the regulation of potent cellular functions (including self-renewal, proliferation, and cell regeneration). As the most representative signaling protein of the Wnt family, it is widely distributed and plays a key role in the regulation of pleiotropic cellular functions, including self-renewal, proliferation, differentiation, and motility.Wnt3a is an important ecological niche component for maintaining the proliferation of Lgr5-positive stem cells in intestinal epithelial cells.

Currently, Wnt3a protein has been used in organoid cultures such as intestinal, gastric, pancreatic, hepatic, thyroid and ovarian cancers, etc. Its high activity, batch-to-batch stability and serum-free preparation are the keys to the success of organoid experiments. Wnt3a protein purified by NextSense is generally used in organoid culture at a working concentration of 30-100 ng/mL, which is usually able to activate the Wnt signaling pathway and promote organoid development and cell function. The optimal concentration to use needs to be optimized and determined based on the specific experimental purpose, cell type and culture conditions. This product is supplied in liquid form and is characterized by high purity, high activity, low endotoxin and no animal-derived components, and has been validated in multiple organoid cultures.

Compounding Methods

Brief centrifugation before opening is recommended to bring the contents to the bottom. Reconstitute to a concentration of 0.1-1.0 mg/mL in sterile distilled water. The stock solution should be divided into aliquots and stored at -20°C.
