Beschreibung
B-cell activating factor (BAFF) is a TNF ligand superfamily member and has been designated TNFSF13B. Produced by macrophages, dendritic cells, and T lymphocytes, BAFF promotes the survival of B cells and is essential for B cell maturation (1-4). BAFF binds to three TNF receptor superfamily members: B-cell maturation antigen (BCMA/TNFRSF17), transmembrane activator and calcium-modulator and cyclophilin ligand interactor (TACI/TNFRSF13B) and BAFF receptor (BAFF R/BR3/TNFRSF13C). These receptors are type III transmembrane proteins that lack a signal peptide. Whereas TACI and BCMA bind BAFF and another TNF superfamily ligand, APRIL (a proliferation-inducing ligand), BAFF R selectively binds BAFF. The BAFF R extracellular domain lacks the TNF receptor canonical cysteine-rich domain (CRD) and contains only a partial CRD with four cysteine residues. Human and mouse BAFF R share 56% aa sequence identity. BAFF R is highly expressed in spleen, lymph node and resting B cells. It is also expressed at lower levels in activated B cell, in resting CD4+ T cells, in thymus and peripheral blood leukocytes.

Source |
Recombinant Human BAFFR/TNFRSF13C Protein is expressed from HEK293 Cells with His tag at the C-terminal. It contains Ser7-Ala71. |
Molecular Weight |
The protein has a predicted MW of 7.6 kDa. Due to glycosylation, the protein migrates to 13-25 kDa based on Tris-Bis PAGE result. |
Physical Appearance |
Sterile Filtered White lyophilized (freeze-dried) powder. |
Purity |
> 95% as determined by SDS-PAGE |
Activity |
1.ELISA Data: Immobilized Human BAFFR, His Tag at 0.5μg/ml (100μl/Well). Dose response curve for Human BAFFR Antibody, hFc Tag with the EC50 of 6.0ng/ml determined by ELISA. 2.ELISA Data: Immobilized Human BAFF (Trimer) , hFc Tag at 5μg/ml (100μl/Well) on the plate. Dose response curve for Human BAFFR, His Tag with the EC50 of 2.3μg/ml determined by ELISA. 3.SPR Data: Human BAFFR, His Tag captured on CM5 Chip via anti-his antibody can bind Human BAFF Trimer, hFc-Flag Tag with an affinity constant of 1.21 nM as determined in SPR assay (Biacore T200). |
Endotoxin |
< 1.0 EU per 1μg of the protein by the LAL method. |
Formulation |
Lyophilized from 0.22 μm filtered solution in PBS (pH 7.4). Normally 5% trehalose is added as protectant before lyophilization. |
Reconstitution |
Centrifuge tubes before opening. Reconstituting to a concentration more than 100 μg/mL is recommended (usually we use 1 mg/mL solution for lyophilization). Dissolve the lyophilized protein in distilled water. |
Storage
Transport with ice packs. Store at -20℃ to -80℃, valid for one year.
Once reconstituted, store unopened at -20 to -80°C for 3 to 6 months. After reconstitution, store at 2-8°C for 2-7 days.
It is recommended to store in aliquots and freeze for the first use to avoid repeated freezing and thawing.
Note
1. Avoid repeated freezing and thawing.
